High number of pathogenic variants in non-UKBB individuals

Thank you for your work on the new gnomAD v4.0 version.

I work in the field of rare diseases, and recently we noticed that some known pathogenic variants were present with high frequency in the new version of gnomAD.

As an example, I looked at the p.Phe508del mutation in the CFTR gene, which is found at the homozygous state in 58 individuals in gnomAD V4, and 1 individual in gnomAD V2. I first thaught that these individuals were taken from UKBB, but in the vcf file it appeared that they were categorized as non-ukb.

Is it possible that non-ukb individuals could have such a rare disease?

Hi Kevin,

While gnomAD removes cohorts recruited for severe pediatric disease, the database does contain individuals with rare disease because gnomAD contains data from large biobanks that recruit from the general population. A list of projects that contributed data to gnomAD is available here and a table containing the percent of individuals with certain common diseases can be found here.
